Abstract
Fast dissolving tablets (FDTs) have emerged as an advanced oral drug delivery system designed to improve patient compliance, especially in pediatric, geriatric, and dysphagic patients who experience difficulty in swallowing conventional tablets and capsules. The present study focuses on the formulation and evaluation of fast dissolving tablets of Paracetamol using suitable superdisintegrants by direct compression method. Paracetamol is widely used as an analgesic and antipyretic drug, but its conventional dosage forms may show delayed onset of action and swallowing difficulties in certain patient groups. Therefore, the development of fast dissolving tablets offers a better alternative for rapid drug release and enhanced therapeutic efficacy. In this study, different concentrations of superdisintegrants such as Sodium Starch Glycolate and Croscarmellose Sodium were incorporated into the formulation. The prepared tablets were evaluated for both pre-compression and post-compression parameters including bulk density, tapped density, angle of repose, Carr’s index, Hausner ratio, hardness, thickness, friability, weight variation, disintegration time, wetting time, drug content uniformity, and in-vitro dissolution studies. The results demonstrated satisfactory flow properties of powder blends and acceptable tablet characteristics. The tablets showed uniform weight, sufficient hardness, low friability, rapid wetting time, and quick disintegration. Among all formulations, formulation F7 exhibited the best performance with minimum disintegration time and more than 95% drug release within 20 minutes. The increase in concentration of superdisintegrants significantly reduced disintegration time and enhanced drug release. The study concludes that fast dissolving tablets of Paracetamol can be successfully formulated using direct compression technique and suitable superdisintegrants. These formulations provide rapid onset of action, improved patient convenience, and enhanced compliance, making them a promising alternative to conventional oral dosage forms.
